RESUMO:

Brazilian poultry is prominent in the production and exportation of chicken meat, being
undoubtedly important to detect undesirable pathogens in the production chain. From
this perspective, the presence of Salmonella sp. in poultry farming is of concern as it
interferes with the consumer market as it is one of the major microorganisms involved in
foodborne diseases. In order to seek alternatives in reducing bird contamination,
Normative Instruction N o 20 of October 21 st 2016 of the Ministry of Agriculture, Livestock
and Supply (MAPA), aims to control and monitor Salmonella sp. in commercial poultry
and slaughter establishments, requiring boot sock laboratory tests a few days before
slaughter, which continue to be carried out in accordance with Ordinance 126 of
November 3 rd 1995 (MAPA). In order to expedite the detection process, this study aims
to optimize the 3M TM Molecular Detection Assay 2: Salmonella (MDS ® ), according to
Ordinance 126, for detection of Salmonella sp. in boot sock moistened with 1% peptone
water. For the development of the methodology, ISO 16140-2: 2016 was followed, and
200 boot sock samples collected from poultry broilers from the west of Santa Catarina
were analyzed for each reference protocol (MDS ® Protocol, with Tetrathionate Broth
(A)) and alternative (Brain Heart Broth - BHI (B)), obtaining confirmatory test with the
traditional methodology (Ordinance 126). From the 200 samples processed in protocol
A, 23 samples were actually positive for Salmonella sp., 167 negative, 1 false positive
and 9 false negative, while in protocol B there were 24 positive, 175 negative, 0 false
positive and 1 false negative. Therefore, in protocol A, sensitivity of 72% and specificity
of 99% were obtained; and in protocol B 96% sensitivity and 100% specificity.
