Dairy cattle are one of the main branches of animal production in the country. Leptospirosis is among the most common reproductive disease in cattle and causes great economic losses to the activity. Not being a disease of compulsory notification in Brazil, there is difficulty in understanding the extent of infection in any part of the country. As the bacteriological culture is laborious and presents low sensitivity, it was aimed to detect, by means of molecular technique, the presence of Leptospira spp. in cattle in the region of Concórdia, estimating its prevalence and relating it to risk factors. Samples were collected from 178 animals in slaughterhouse. Blood was collected at time of bleeding, urine was collected by bladder puncture, and kidney tissue samples were collected in duplicate after inspection, one packed on ice and one sample in formaldehyde. Formalized renal tissue samples were embedded in paraffin for posterior histology with silver staining of the positive samples in the Polymerase Chain Reaction (PCR) technique. Serum samples were frozen for future serotyping of the positive samples in a specialized laboratory. Renal tissue and urine were frozen for PCR. At the moment, the techniques of extracting DNA from urine and renal tissue samples are being standardized by the phenol-chloroform method. PCR will be performed with the primers previously described (Lepto-2-F/Lepto-504-R and P24-F/Lepto-P23-R). An epidemiological questionnaire was elaborated to trace a profile of the animal's origin farms and its application is being started. The expectation is to obtain a more accurate diagnosis of the reproductive problems in the region, with an applicable methodology and the objective of guiding the producers on the control and prevention of infection, also contributing with new epidemiological data of the studied area.
