SUMMARY:

With the intensification of aquaculture, routine management practices such as biometrics, blood and semen collection, hormonal implants, oocyte extrusion, and transportation expose fish to stressful factors. These manipulations often induce physiological stress responses that can lead to immunosuppression and growth retardation. To reduce these adversities, anesthetics are used to mitigate the physiological stress responses. Several studies have confirmed the effectiveness of using essential oils as anesthetics in fish, therefore, the aim of this study is to evaluate the ideal concentration and its effects on stress responses in Oreochromis niloticus subjected to an anesthesia protocol using Cinnamomum zeylanicum essential oil. Experiment I: Evaluation of different concentrations of C. zeylanicum essential oil as an anesthetic. One hundred fish (10 per concentration) will be randomly distributed into 10 treatments, which include nine concentrations of cinnamon essential oil (20, 30, 40, 50, 60, 70, 80, 90, and 100 μl/L), and one control group subjected only to the highest volume of the diluent used (ethyl alcohol). Experiment II: Evaluation of physiological parameters (cortisol, glucose, lactate, hemoglobin, and hematocrit) exposed to the ideal concentration of essential oil determined in Experiment I. Twenty fish will be used (10 controls and 10 in the treatment group). With this experiment, it is expected to validate the application of C. zeylanicum essential oil as an anesthetic for tilapia farming and determine the ideal concentration for use. The experiments will be conducted in november 2023 at the Aquaculture Laboratory, Federal Institute of Education, Science and Technology Catarinense, Araquari, Brazil.
