SUMMARY:

Bovine Euritrematosis is caused by Eurytrema coelomaticum, a trematode of pancreatic ducts in ruminants, predominantly in the southern states of Brazil. Macroscopic pancreatic lesions were related to pancreatic atrophy, fibrous interstitial pancreatitis and total or partial obliteration of the pancreatic ducts. It is conjectured that they may cause disorders in the secretory functions of the pancreas, digestive and metabolic processes dependent on these, causing losses of production in dairy and beef cattle. There is not effective clinical treatment for the control of bovine Euritrematosis and only the drug praziquantel has been proven effective in vitro. The objective of this work is to evaluate, separately, the anthelmintic activity of the drugs praziquantel and nitazoxanide against the adult parasite Eurytrema coelomaticum. Specimens of E. coelomaticum will be obtained from the pancreas of naturally infected cattle, collected from animals slaughtered in slaughterhouses in the western region of Santa Catarina. The parasites will be kept in saline solution and placed in a thermal box with a temperature of 37ºC and sent to the Animal Parasitology laboratory of the Federal Catarinense Institute - Campus Concórdia. Subsequently, they will be washed in RPMI 1640 buffered with 20µM HEPES for complete removal of the host's blood. The anatomical integrity of the parasites will be evaluated under an optical microscope and intact specimens will be transferred to tissue culture plates containing 2 mL of RPMI 1640 medium supplemented with penicillin (50UI / mL), streptomycin (50 mg / mL) and fetal bovine serum ( 10%). 10 helminths will be used per plate, divided into: Group I - Control; Group II - Praziquantel treatment; Group III - Treatment with Nitazoxanide; in three repetitions. As a control, the RPMI medium will be used without antiparasitic drugs. The culture plates will be incubated at 37 ºC with 5% CO2 and examined every hour until the first 12 hours of the experiment, and after, every 12 hours, totaling 72 hours of incubation. During this period, the viability of helminths will be checked, observing the motility and the presence of parasite eggs in the culture medium. Motility will be scored according to the following criteria: 3 (move the whole body), 2 (move only parts of the body), 1 (immobile, but not dead, without stains of vital dye) and 0 (immobile, stained with vital dye) . After 72 hours of incubation, the parasites will be sent for classical histological processing to assess tissue damage. The results obtained will be found in the Kruskal-Wallis test by statistical analysis performed with the computer program SAS® (Statitical Analysis System) and determined when P <0.05.
